SCOTTSDALE, Ariz. August 29, 2012 A Phase I/II, multi-center trial designed to test the safety and preliminary efficacy of a first in class cancer treatment opened worldwide today at the Virginia G. Piper Cancer Center Clinical Trials at Scottsdale Healthcare, a partnership between Scottsdale Healthcare and the Translational Genomics Research Institute (TGen). PR610 is designed to become activated in tissues with low levels of oxygen, which is characteristic of many cancers. This "targeted" approach should deliver more active drug to cancer tissue and less active drug to normal tissue.

The two-part trial, sponsored by the drug's manufacturer, Proacta Inc., will first determine acceptable dose levels of PR610 in patients with various types of advanced cancer. After determining acceptable dose levels, the study will evaluate the safety and efficacy of PR610 when given to patients with non-small cell lung cancer whose tumors contain a certain genetic mutation. Other goals of the study are to measure levels of PR610 in the blood.

"PR610 is a promising and innovative compound that merges delivery of a drug to the cancerous, oxygen depleted tissue and it also targets the communication functions that govern the growth and behavior that drive certain cancers," said Dr. Glen Weiss, Clinical Associate Professor at TGen and Director of Thoracic Oncology at Virginia G. Piper Cancer Center Clinical Trials at Scottsdale Healthcare.

Research has shown that some cancer tissue has lower levels of oxygen than normal tissue. This lower level of oxygen appears to make the cancer tissue more resistant to radiation and chemotherapy, which allows the cancer to become more aggressive and spread. PR610 is designed to become active in cancer tissue with low levels of oxygen.

"Proacta is very excited to be working with Virginia G. Piper Cancer Center Clinical Trials and TGen on this important study. We purposely limited participation in this study to those cancer centers with extensive experience and expertise in conducting clinical oncology trials," said Dr. John Gutheil, CEO of Proacta.

Non-small cell lung cancer is a disease in which cancerous cells form in the tissues of the lung. There are several types, and chances of recovery are determined by many factors, such as smoking and lifestyle. Unfortunately, current treatments options do not cure the majority of patients with non-small cell lung cancer.

About 44 people will participate in this study at research sites in the U.S. and New Zealand, with the Virginia G. Piper Cancer Center at Scottsdale Healthcare being the first to enroll a patient.

Harvard scientists have, for the first, time created a type of "cyborg" tissue by embedding a three-dimensional network of functional, bio-compatible nanoscale wires into engineered human tissues.

As described in a paper published August 26 in Nature Materials, a multi-institutional research team led by Charles M. Lieber, the Mark Hyman, Jr. Professor of Chemistry at Harvard and Daniel Kohane, a Harvard Medical School professor in the Department of Anesthesia at Children's Hospital Boston developed a system for creating nanoscale "scaffolds" which could be seeded with cells which later grew into tissue.

Also contributing to the work were Robert Langer, from the Koch Institute at the Massachusetts Institute of Technology, and Zhigang Suo, the Allen E. and Marilyn M. Puckett Professor of Mechanics and Materials at Harvard's School of Engineering and Applied Sciences.

"The current methods we have for monitoring or interacting with living systems are limited," said Lieber. "We can use electrodes to measure activity in cells or tissue, but that damages them. With this technology, for the first time, we can work at the same scale as the unit of biological system without interrupting it. Ultimately, this is about merging tissue with electronics in a way that it becomes difficult to determine where the tissue ends and the electronics begin."

The research addresses a concern that has long been associated with work on bioengineered tissue ? how to create systems capable of sensing chemical or electrical changes in the tissue after it has been grown and implanted. The system might also represent a solution to researchers' struggles in developing methods to directly stimulate engineered tissues and measure cellular reactions.

"In the body, the autonomic nervous system keeps track of pH, chemistry, oxygen and other factors, and triggers responses as needed," Kohane explained. "We need to be able to mimic the kind of intrinsic feedback loops the body has evolved in order to maintain fine control at the cellular and tissue level."

Using the autonomic nervous system as inspiration, Bozhi Tian, a former doctoral student under Lieber and former postdoctoral fellow in the Kohane and Langer labs, and collaborator Jia Liu worked in Lieber's lab at Harvard to build mesh-like networks of nanoscale silicon wires?about 30 - 80 nm in diameter?shaped like flat planes or in a reticular conformation.

The process of building the networks, Lieber said, is similar to that used to etch microchips.

Beginning with a two-dimensional substrate, researchers laid out a mesh of organic polymer around nanoscale wires, which serve as the critical nanoscale sensing elements. Nanoscale electrodes, which connect the nanowire elements, were then built within the mesh to enable nanowire transistors to measure the activity in cells without damaging them. Once complete, the substrate was dissolved, leaving researchers with a net-like sponge or a mesh that can be folded or rolled into a host of three dimensional shapes.

Once complete, the networks were porous enough to allow the team to seed them with cells and encourage those cells to grow in 3D cultures.

"Previous efforts to create bioengineered sensing networks have focused on two-dimensional layouts, where culture cells grow on top of electronic components, or on conformal layouts where probes are placed on tissue surfaces," said Tian. "It is desirable to have an accurate picture of cellular behavior within the 3D structure of a tissue, and it is also important to have nanoscale probes to avoid disruption of either cellular or tissue architecture."

Using heart and nerve cells, the team successfully engineered tissues containing embedded nanoscale networks without affecting the cells' viability or activity. Using the embedded devices, they were able to detect electrical signals generated by cells deep within the tissue, and to measure changes in those signals in response to cardio- or neuro-stimulating drugs.

Researchers were also able to construct bioengineered blood vessels, and used the embedded technology to measure pH changes ? as would be seen in response to inflammation, ischemia and other biochemical or cellular environments ? both inside and outside the vessels.

Though a number of potential applications exist for the technology, the most near-term use, Lieber said, may come from the pharmaceutical industry, where researchers could use the technology to more precisely study how newly-developed drugs act in three dimensional tissues, rather than thin layers of cultured cells. The system might also one day be used to monitor changes inside the body and react accordingly, whether through electrical stimulation or the release of a drug.

Llama semen is loaded with NGF, says study coauthor and veterinarian Gregg Adams of the University of Saskatchewan in Saskatoon, Canada. The protein is also found in the semen of bulls and humans.

"If we find that NGF is also effective in women, it will obviously have huge implications for treating male infertility conditions,? says reproductive biologist Raj Duggavathi of McGill University?s campus in Sainte-Anne-de-Bellevue, Canada. ?It could be a big boost for couples.

NGF is well-known to biologists but not in the context of reproduction. Nerve cells typically spit the protein out to tell neighboring cells to grow.

Previous findings that semen could trigger ovulation in llamas challenged conventional wisdom that the physical movements of sex were what stimulated egg drop in the animals, says Adams. Unlike humans cows, horses and sheep, which ovulate on a regular cycle, some animals rely on a little action to get their eggs moving.

Adams? team had shown that semen ? filtered free of sperm ? injected into llamas? leg muscles could set off ovulation all on its own ? no bump-and-grind required. Semen from stallions, cattle, boars and rabbits could also prod llamas to ovulate.

Adams? team confirmed that the llama semen substance was NGF by using a technique usually seen in fertility clinics: injections followed by ultrasounds. The researchers gave llamas shots of either the purified semen substance or NGF from mouse salivary glands ? a classic source of the protein. Then, they checked the animals? reproductive organs with daily ultrasounds. Both substances made ovulation rates shoot up.

I?m fairly convinced? that NGF is the ovulation-sparking semen substance, says reproductive biologist Bruce Murphy of the University of Montreal. ?It?s a blockbuster paper,? he says. The work is the first to link NGF to reproductive function.

Next, Adams? team plans to investigate NGF?s role in human semen ? to see if it?s connected to fertility in people as well as llamas. Because women, unlike llamas, don?t need semen to ovulate, it?s not clear yet if NGF is also important for people, says reproductive biologist Dan Bernard of McGill University in Montreal. ?It?s still early.? Meet Kevin and Caroline, two of the owners of the gorgeous homewares, gift and furniture shops ? or for want of a better word, ?lifestyle shops?. Interviewing these two is really like talking to retail royalty! Kevin and Caroline have had a number of clothing, homewares and shoe stores over the years, and at one stage probably made up a large part of the market of trendy Perth stores. Now they?re threatening to do it all over again with the beautiful Store and Corner Store concepts popping up all over town, and really, we couldn?t be more delighted!

We love a family business! Who is behind Corner Store?

So do we! There?s Kevin and Caroline, Annie and Adam (Caroline?s brother) and all our wonderful staff.

Do you think family and business mix well?

We have to say yes, the other two might read this! But truthfully it?s great. We have our moments but when you are bound together by more important things in life it makes business a breeze.

What are each of your strengths in the business?

Caroline and Annie do the buying, merchandising and the general running of the day to day shop floor stuff. Kevin does the accounts, wages etc and works creatively with Kylie, whilst Adam is ?logistics?, organising furniture deliveries, manufacturing as well as head barista. Having said that the lines are often blurred as there is much crossover.

My early teens were spent browsing the second hand threads in Lush, when my niece was born I gushed over the baby toys in Store, and presently I?m swooning over the furniture in your showrooms, daydreaming about filling my house with the pieces. You?ve been a force of style in my life for the best part of 2 decades! Can you tell us a bit about the history of your retail stores and what permutations of stores you?ve been through?

Oh my! That does make us feel old! But it has been a progression in our lives too.

Lush started in 1992 as a market stall in the cultural centre when Caroline made clothes and sold them. We mixed in vintage clothes and opened our first shop on Cantonment Street Fremantle in ?94. Lush turned to Lick in ?96. After many trips to Europe gathering vintage clothing and ephemera we noticed a move to ?lifestyle? shops where all needs were met. We thought we?d have a go and with Annie and Adam set up Store in ?99 on Market Street just after the arrival of our first borns which tied in nicely with our stock mix too.

Lick moved in next to Store in 2001, which was a busy year as we opened up Wanderlust Footwear on Cantonment St with our friend Sarah Wallwork. We had quite a nice little triangle going on there for a while. Store Claremont opened in 2005 on St Quentin Ave, followed by Mount Lawley in 2008 which also saw the birth of Corner Store on Stirling Highway Claremont and the death of Store on St Quentin at the hands of a Hawaiian/Multiplex wrecking ball.

Last year it seemed like all we did was renovate. The original Store moved next door into the lovely old Lick shop and after months of throwing white paint around, Corner Store on South Terrace opened in October? I feel like a lie down now!

How do you keep it so fresh over the years?

What is the best thing about your job?

It?s not a job!

Take us through a typical day of your work:

The days vary but generally we start very early to get ahead before we get caught up in normal family life: preparing breakfasts, lunches and school runs. Once that?s done we then do our rounds of the shops, dropping off stock, making deliveries to customers, catch up with our staff and just general grown up business stuff. We then have a period after school pick ups where we work from home but we don?t really have that Work/Home separation as it?s so much a part of our lives!

What have been the hardest hurdles to face over the years in the retail biz?

I remember once the CD player broke and I had to sing to every customer that came in, people can be so cruel? That was a tough day.

Which local creatives do you admire?

Kylie Thevenau, our brilliant designer and ?family? member who embodies our style and ethos so well. And you two too!

Any tips for those hoping to open a shop of their own?

Don?t!

Well, don?t spend too much on the fit-out ? white paint covers a multitude of sins. So many businesses have folded after investing so much money on the shell ? it?s all about what you are selling. It?s a bit like divorcing after an elaborate wedding. Keep it simple.

iPad 2 CDMA front panel

Just replaced my panel, instructions to take panel off were ok but...There are some steps that need to be included in the reassembly namely:

1.) Simple instructions to reattach button and front camera bracket. A picture is worth a thousand words, I was not sure if I had the button rotated in the right direction.

2.) Correct order to replace parts: I pulled out the LCD to clean up some broke glass. With the display out I affixed the adhesive strips only to discover I had to remove the corners to get to the screws.

3.) Adhesive strips: there needs to be a step showing were all the various strips go. Additionally they are backwards (in my opinion) if you want to adhere them to the body you have to first pull both sides of the adhesive off. It would be nice if they were flipped over on the paper but I know this is an OEM product and not your fault. A caution note would have saved me a few f-bombs.

Mitt Romney attacked President Barack Obama Saturday over the future of Medicare in his first weekly podcast ahead of the November election.

In the address posted on his website, the GOP's presidential candidate said Obama?s healthcare law had taken $716 billion from the fund to finance ?his takeover of the healthcare system.?


?Now if that wasn?t bad enough, his healthcare law also put in place a board of 15 unelected bureaucrats and gave them the power to make additional cuts to Medicare without even having to get approval from Congress,? he said.

?This means they could deny elderly Americans the care they?ve worked for their entire lives -- all because President Obama trusts bureaucrats more than he trusts seniors and their doctors,? he said.

?And here?s one more troubling aspect of all this: According to independent, non-partisan scorekeepers, these cuts the President?s people will take to Medicare won?t prevent it from going bankrupt: Experts estimate that Medicare?s trust funds will be exhausted just twelve short years from now,? Romney added.

The Democrats charge that Mitt Romney and Ryan would gut programs for older Americans such as Medicare and Social Security.?
Obama plans to dig in on that point in New Hampshire on Saturday with stops in Windham and Rochester. Aides say he will cast voters' choice as one between two very different approaches to government, The Associated Press reported.

The Obama campaign has claimed Romney's running mate Paul Ryan was forced to "flip flop" on Medicare by the GOP candidate. And Rep. Chris Van Hollen (D-MD) said the Romney-Ryan plan would "hasten the insolvency of the Medicare program by 8 years, that's according to the Medicare trustees."

On the campaign trail this week, Romney said the?Romney-Ryan plan to transform Medicare was "very similar" to Medicare Advantage.

But?Marsha Gold, a health-care expert at Mathematica Policy Research, said the big difference between the two is this: Medicare Advantage, like Medicare, is a defined benefit -- where the government guarantees a level of coverage -- while the Romney-Ryan plan transforms Medicare into a defined contribution -- where the government decides what it will pay.

In the podcast Saturday, Romney reiterated a pledge to repeal ?Obamacare? ? that he characterized as ?threatening seniors? and ?a maze of new federal mandates, and taxes, and penalties that?s hampering job creation? ? and work on ?real solutions to save Medicare.?

Romney added that the plan would not change Medicare for those who are retired or ?near? retirement.

For ?younger Americans,? he said future retirees would be provided with federal financial support and would be able to choose from a list of Medicare-approved coverage plans, including a traditional Medicare option.

?The amount of financial support that a person would get would be adjusted based on their income; more help would go to the poor or the sick -- and less help would go to those that are financially better off,? Romney added.

?It would be based on how much the plans cost so that seniors always have access to affordable, quality coverage. And no senior could ever be denied coverage for any reason,? he said.
